ABSTRACT

Here, quercetin-mediated silver nanoparticle (AgNP) formation combined with loop-mediated isothermal amplification (LAMP) was introduced to colorimetrically detect two major infectious pathogens, SARS-CoV-2 and Enterococcus faecium, using a foldable PMMA microdevice. The nitrogenous bases of LAMP amplicons can readily form a complex with Ag+ ions, and the catechol moiety in quercetin, which acted as a reducing agent, could be chelated with Ag+ ions, resulting in the easy electron transfer from the oxidant to the reductant and producing brown-colored AgNPs within 5 min. The introduced method exhibited higher sensitivity than agarose gel electrophoresis due to more active redox centers in quercetin. The detection limit was attained at 101 copies µL-1 and 101 CFU mL-1 for SARS-CoV-2 RNA and E. faecium, respectively. A foldable microdevice made of two pieces of PMMA that fully integrates DNA extraction, amplification, and detection processes was fabricated to establish practical applicability. On one PMMA, DNA extraction was performed in a reaction chamber inserted with an FTA card, and then LAMP reagents were added for amplification. Silver nitrate was added to the reaction chamber after LAMP. On the other PMMA, quercetin-soaked paper discs loaded in the detection chamber were folded toward the reaction chamber for colorimetric detection. An intense brown color was produced within 5 min when heated at 65 °C. The introduced colorimetric assay, which is highly favorable for laboratory and on-site applications, could be a valuable alternative to conventional methods for detecting infectious diseases, given its unique principle, simplicity, and naked-eye detection.

ABSTRACT

OBJECTIVES: There have been limited data on the risk of onward transmission from individuals with Omicron variant infections who return to work after a 5-day isolation. We evaluated the risk of transmission from healthcare workers (HCWs) with Omicron variant who returned to work after a 5-day isolation and the viable virus shedding kinetics. METHODS: This investigation was performed in a tertiary care hospital, Seoul, South Korea. In a secondary transmission study, we retrospectively reviewed the data of HCWs confirmed as COVID-19 from March 14 to April 3, 2022 in units with 5 or more COVID-19-infected HCWs per week. In the viral shedding kinetics study, HCWs with Omicron variant infection who agreed with daily saliva sampling were enrolled between February and March, 2022. RESULTS: Of the 248 HCWs who were diagnosed with COVID-19 within 5 days of the return of an infected HCW, 18 (7%) had contact with the returned HCW within 1 to 5 days after their return. Of these, 9 (4%) had an epidemiologic link other than with the returning HCW, and 9 (4%) had contact with the returning HCW, without any other epidemiologic link. In the study of the kinetics of virus shedding (n=32), the median time from symptom onset to negative conversion of viable virus was 4 days (95% CI, 3 to 5 days). CONCLUSIONS: Our data suggest that the residual risk of virus transmission after 5 days of isolation following diagnosis or symptom onset is low.

ABSTRACT

The sudden adoption of working from home (WFH) during the COVID-19 pandemic has required the reconfiguration of home spaces to fit space for remote work into existing spaces already filled with other domestic functions. This resulted in blurring of home and work boundaries, the potential lack of space for telecommuting from home, and telecommuters' feelings of crowding. Numerous studies have shown the negative effects of crowding feelings on workers' responses. This study focused on the issue of crowding in the residential workspace. An online survey was conducted to investigate how features of the home workspace correlate with telecommuters' feelings of crowding and how these feelings affect satisfaction, health, and productivity. As a result, we found that various environmental features of home workspaces (e.g., house size, purpose of workspace, accessible balcony, lighting, noise, etc.), as well as psychological aspects (e.g., individual control over space use), had significant effects on telecommuters' feelings of crowdedness. It was also found that feelings of crowding in the WFH environment can directly and indirectly affect teleworkers' satisfaction with work environments, well-being, and work performance. Based on the results, we offered various potential ways to alleviate overcrowding issues in the WFH context.

ABSTRACT

Today, collaborative playlists (CPs) translate long-standing social practices around music consumption to enable people to curate and listen to music together over streaming platforms. Yet despite the critical role of CPs in digitally connecting people through music, we still understand very little about the needs and desires of real-world users, and how CPs might be designed to best serve them. To bridge this gap in knowledge, we conducted a survey with CP users, collecting open-ended text responses on what aspects of CPs they consider most important and useful, and what they viewed as missing or desired. Using thematic analysis, we derived from these responses the Codebook of Critical CP Factors, which comprises eight categories. We gained insights into which aspects of CPs are particularly useful-for instance, the ability for multiple collaborators to edit a single playlist-and which are absent and desired-such as the ability for collaborators to communicate about a CP or the music contained therein. From these findings we propose design implications to inform further design of CP functionalities and platforms, and highlight potential benefits and challenges related to their adoption in current music services.
